Descripción: The idea that memories are immutable after consolidation has been challenged. Several reports have shown that after the presentation of a specific reminder, reactivated old memories become labile and again susceptible to amnesic agents. Such vulnerability diminishes with the progress of time and implies a re-stabilization phase, usually referred to as reconsolidation. To date, the main findings describe the mechanisms associated with the labilization-reconsolidation process, but little is known about its functionality from a biological standpoint. Indeed, two functions have been proposed. One suggests that destabilization of the original memory after the reminder allows the integration of new information into the background of the original memory (memory updating), and the other suggests that the labilization-reconsolidation process strengthens the original memory (memory strengthening). We have previously reported the reconsolidation of human declarative memories, demonstrating memory updating in the framework of reconsolidation. Here we deal with the strengthening function attributed to the reconsolidation process. We triggered labilization-reconsolidation processes successively by repeated presentations of the proper reminder. Participants learned an association between five cue-syllables and their respective response-syllables. Twenty-four hours later, the paired-associate verbal memory was labilized by exposing the subjects to one, two or four reminders. The List-memory was evaluated on Day 3 showing that the memory was improved when at least a second reminder was presented in the time window of the first labilization-reconsolidation process prompted by the earlier reminder. However, the improvement effect was revealed on Day 3, only when at least two reminders were presented on Day2 and not as a consequence of only retrieval. Therefore, we propose central concepts for the reconsolidation process, emphasizing its biological role and the parametrical constrains for this function to be operative. © 2011 Forcato et al.

Descripción: The fluorescence emission of merocyanine 540 (MC540) in 95% ethanol was studied under continuous irradiation. Fluorescence spectra from excited states of both normal (N) and photoisomeric (P) species are identical. The laser fluence dependence of the fluorescence intensity is interpreted on the basis of a photochromic system involving N and P ground states and first excited singlet states. Common flash photolysis equations are generalized in order to include a photoequilibrium between isomers. The emission data are used together with previous flash photolysis and optoacoustic results to obtain P fluorescence and photoisomerization quantum yields as 0.07 ± 0.02 and 0.20 ± 0.04, respectively, P absorption cross section at the maximum (560 nm) as 4.74 × 10-16 cm2 (125 × 103 M-1·cm-1), and the energy difference between the ground states as 165 kJ·mol-1. Internal conversion, which can take place as a direct process or through a twisted state, is the main deactivation pathway for excited singlet P. © 1990 American Chemical Society.

Descripción: In the region of the Main Andes of San Juan, exists a high concentration of landslide deposits that can originate natural dams. The outburst of these dams generated by slide is a common process in this morphology. The analyses of satellite imagery and air photos of previous years allow reconstructing the characteristics and the origin of the slides that formed those natural dams. In this region of the Main Andes, the rapid uplift and erosion of the mountain chain has created abrupt slopes and an internal structure in the materials that determined an increase of the susceptibility to the slope collapse without triggers as the seismic activity or precipitations. The objectives of the present work are (1) to analyze the causes that formed the natural dam and produced the landslide dam and development of the Los Erizos lagoon in the middle course of the Santa Cruz river; (2) the outburst of the dam and their consequent flood; and (3) the morphologic characteristic of the study region. The temporary comparison between air photos with satellite imageries indicates that the natural dam was generated by slip of the western hillside of the Santa Cruz mountain range, located below the Cerro Estrella. However, the constant water flow to the lagoon, has increased its water level until reaching the maximum benchmark of the dam, produced the beginning of the lake drainage and weakening of the dam causing the outburst and its consequent flood. Based on satellite imagery, a temporary analysis of the lagoon is presented, indicating that monitoring is an important tool to alert in the whole San Juan river basin for possible natural dikes, preventing hazards to the populations down water. The alluvium of November 12 of 2005, discharged 32.100.000 m3 in 67 minutes and traveled 254 km in less than 12 hours.

Descripción: A significant wave activity (WA) in the upper troposphere and lower stratosphere, mainly during winter, was detected at midlatitudes in the southern hemisphere (30-40S) above the Andes Range, from an analysis of Global Positioning System Radio Occultation (GPS RO) temperature profiles retrieved by CHAMP (CHAllenging Mini-satellite Payload) and SAC-C (Satélite de Aplicaciones Científicas-C) Low Earth Orbit (LEO) satellites, between May 2001 and February 2006. The possible main gravity wave sources in this region are: i) orographic forcing, ii) geostrophic adjustment and iii) deep convection. The available vertical resolution of GPS RO soundings does not rule out any of these alternatives. Based on satellite imaginary, the WA enhancements cannot be attributed to deep convection events. Inertia-gravity waves (IGWs) could be generated after a geostrophic adjustment process, following a perturbation of the zonal jet situated above the Andes Mountains by mountain waves (MWs). The monthly WA intensity follows the zonal wind velocity strength according to its seasonal variability at jet altitudes. As the GPS-LEO lines of sight are roughly meridionally aligned and the morphology of the Andes at middle latitudes is predominantly north-south, it was possible to detect MWs as well as IGWs from GPS RO temperature profiles. This characteristic does not apply for other mountain range alignments. From the analysis of a numerical simulation at the time and location of a single RO event with very strong WA, two main modes of oscillation with horizontal wavelength around 40 and 200 km were identified. Descripción: The Mediator complex is a greater than 1-megadalton complex, composed of about 30 subunits and found in most eukaryotes, whose main role is to transmit signals from DNA-bound transcription factors to RNA Polymerase II. The proteasome is emerging as an important regulator of transcription during both initiation and elongation. It is increasing the number of cases where the proteolysis of transcriptional activators by the proteasome activates their function. This counterintuitive phenomenon was called "activation by destruction." Here, we show that, in Arabidopsis (Arabidopsis thaliana), PHYTOCHROME AND FLOWERING TIME1 (PFT1), the MEDIATOR25 (MED25) subunit of the plant Mediator complex, is degraded by the proteasome and that proteasome-mediated PFT1 turnover is coupled to its role in stimulating the transcription of FLOWERING LOCUS T, the plant florigen, which is involved in the process of flowering induction. We further identify two novel RING-H2 proteins that target PFT1 for degradation. We show that MED25-BINDING RING-H2 PROTEIN1 (MBR1) and MBR2 bind to PFT1 in yeast (Saccharomyces cerevisiae) and in vitro, and they promote PFT1 degradation in vivo, in a RING-H2- dependent way, typical of E3 ubiquitin ligases. We further show that both MBR1 and MBR2 also promote flowering by PFT1- dependent and -independent mechanisms. Our findings extend the phenomenon of activation by destruction to a Mediator subunit, adding a new mechanism by which Mediator subunits may regulate downstream genes in specific pathways. Furthermore, we show that two novel RING-H2 proteins are involved in the destruction of PFT1, adding new players to this process in plants. © 2012 American Society of Plant Biologists.

Descripción: The yeast Yarrowia lipolytica accumulates oils and is able to produce extracellular lipases when growing in different carbon sources including glycerol, the principal by-product of the biodiesel industry. In this study, biomass production of a novel mutant strain of Y. lipolytica was statistically optimized by Response Surface Methodology in media containing biodiesel-derived glycerol as main carbon source. This strain exhibited distinctive morphological and fatty acid profile characteristics, and showed an increased extracellular lipase activity. An organic source of nitrogen and the addition of 1.0 g/l olive oil were necessary for significant lipase production. Plackett-Burman and Central Composite Statistical Designs were employed for screening and optimization of fermentation in shaken flasks cultures, and the maximum values obtained were 16.1 g/l for biomass and 12.2 Units/ml for lipase, respectively. Optimized batch bioprocess was thereafter scaled in aerated bioreactors and the values reached for lipase specific activity after 95 % of the glycerol had been consumed, were three-fold higher than those obtained in shaken flasks cultures. A sustainable bioprocess to obtain biomass and extracellular lipase activity was attained by maximizing the use of the by-products of biodiesel industry.

Descripción: It is universally true in ecological communities, terrestrial or aquatic, temperate or tropical, that some species are very abundant, others are moderately common, and the majority are rare. Likewise, eukaryotic genomes also contain classes or "species" of genetic elements that vary greatly in abundance: DNA transposons, retrotransposons, satellite sequences, simple repeats and their less abundant functional sequences such as RNA or genes. Are the patterns of relative species abundance and diversity similar among ecological communities and genomes? Previous dynamical models of genomic diversity have focused on the selective forces shaping the abundance and diversity of transposable elements (TEs). However, ideally, models of genome dynamics should consider not only TEs, but also the diversity of all genetic classes or "species" populating eukaryotic genomes. Here, in an analysis of the diversity and abundance of genetic elements in >500 eukaryotic chromosomes, we show that the patterns are consistent with a neutral hypothesis of genome assembly in virtually all chromosomes tested. The distributions of relative abundance of genetic elements are quite precisely predicted by the dynamics of an ecological model for which the principle of functional equivalence is the main assumption. We hypothesize that at large temporal scales an overarching neutral or nearly neutral process governs the evolution of abundance and diversity of genetic elements in eukaryotic genomes. © 2013 Serra et al.

Descripción: We present experimental measurements and theoretical predictions of ion transport in agar gels during reversible electroporation (ECT) for conditions typical to many clinical studies found in the literature, revealing the presence of pH fronts emerging from both electrodes. These results suggest that pH fronts are immediate and substantial. Since they might give rise to tissue necrosis, an unwanted condition in clinical applications of ECT as well as in irreversible electroporation (IRE) and in electrogenetherapy (EGT), it is important to quantify their extent and evolution. Here, a tracking technique is used to follow the space-time evolution of these pH fronts. It is found that they scale in time as t1/2 characteristic of a predominantly diffusive process. Comparing ECT pH fronts with those arising in electrotherapy (EChT), another treatment applying constant electric fields whose main goal is tissue necrosis, a striking result is observed: anodic acidification is larger in ECT than in EChT, suggesting that tissue necrosis could also be greater. Descripción: Many image processing applications need real time performance, while having restrictions of size, weight and power consumption. These include a wide range of embedded systems from remote sensing applications to mobile phones. FPGA-based solutions are common for these applications, their main drawback being long development time. In this work a co-design methodology for processor-centric embedded systems with hardware acceleration using FPGAs is applied to an image processing method for localization of multiple robots. The goal of the methodology is to achieve a real-time embedded solution using hardware acceleration, but with development time similar to software projects. The final embedded co-designed solution processes 1600×1200 pixel images at a rate of 25 fps, achieving a 12.6× acceleration from the original software solution. This solution runs with a comparable speed as up-to-date PC-based systems, and it is smaller, cheaper and demands less power. © 2012 Springer-Verlag.

Descripción: A sequence of plutonic mafic rocks inter-stratified with both mafic- and metasedimentary-derived migmatites is found along the San Juan valley in the center of the Sierra de Valle Fértil. This natural example shows the transition from igneous to metamorphic petrologic processes which occurred during the crystallization of mafic magmas and the subsequent partial melting of crystallized gabbroic rocks. This work studies the mineralogical changes associate to this petrologic transition. Thermobarometric estimates based on amphibole-plagioclase indicate that the mafic magmas crystallized at around 1100°C and 5 ± 0.5 kbar. The conditions under which gabbroic rocks were partially melted are estimated using two pyroxenes ther-mometry and amphibole-plagioclase thermobarometry. Similar physical conditions in the range between 740 and 840°C and 5 to 6.5 kbar are recovery from mineral assemblages in the mesosomes and leucosomes of mafic migmatites. The main mineral compositional changes that accompanied the partial process of the gabbroic rocks are: 1) depletion of aluminium content and Mg# ratio in pyroxenes; 2) depletion of anorthite mole fraction of the plagioclases; and 3) depletion of the Mg# ratio in amphiboles. These mineral compositional variations are consistent with those found by experimentally melting mafic protoliths. Experimental results showed that the temperature for promoting amphibole-dehydration melting in mafic proto-liths is of around 850°C, whereas the addition of an aqueous fluid would lower the melting temperature to around 800°C. Combining field relationships, textural features, mineral compositional variations and thermo-barometric estimates with experimental results, we demonstrated that the transition from igneous mafic rocks to mafic migmatites was due to isobaric cooling. Moreover, repetitive intrusion of mafic magmas sustained the sequence at granulite-facies conditions, so avoiding rapid cooling. The natural case from the centre of the Sierra de Valle Fértil is interpreted to expose an example of the plutonic-me-tamorphic core of the Famatinian magmatic arc, where the abnormally high metamorphic gradient provide evidence that large volume of mafic magmas reached and emplaced at paleodepths of between 16 and 20 kilometres.

Descripción: The Payenia basaltic province is a typical retroarc association developed along the foothills of the Andes between 33°40 ́S and 38°00'S. It records two main events, an older, mostly Miocene one (26 to 8 Ma) and a younger Pliocene to Holocene one (younger than 5 Ma). It covers an N-S lowland belt named here the central depression. To the North of 36°20'S, the region was described as Los Huarpes depression, a partially deformed sedimentary basin characterized by a 1000 m-thick, undifferentiated Cenozoic sequence. To the south, Upper Cretaceous to Lower Paleogene layers, instead of the undifferentiated Cenozoic deposits, are covered by basalts. The volcanism additionally covers the western side of the San Rafael Block as well as the fold and thrust belt of the Andean foothills. The Payenia province consists of more than 800 monogenetic basaltic cones, and scarce polygenetic volcanos fed by shallow magmatic chambers. Among the latter, the following examples can be mentioned: Miocene Chachahuén volcano (7 to 5 Ma), composed of andesites, rhyodacites and basalts with high-K and amphibole; Pliocene El Nevado volcano, with calc-alkaline, basaltic trachyandesites, trachytes, dacites and rhyolites; and Upper Pleistocene to Holocene Payún Matrú volcano, composed of trachytes, trachyandesites and trachybasalts. The southernmost area of the central depression is covered by the Pleistocene, Auca Mahuida basaltic shield (1.7 to 0.88 Ma), consisting of trachybasalts, basaltic trachyandesites and trachyandesites. This shield is aligned with Tromen and Domuyo volcanos, defining an NW-SE volcanic belt, oblique to the Andes, whose southern tip corresponds to the Cortaderas lineament. Some of the monogenetic cones are of hydromagmatic origin, whereas a few others exhibit small mantle inclusions. The Payenia retroarc province develops to the south of the Pampean flat slab segment, where the heights of the Andes are smaller. This height difference might have been caused by a higher thermal gradient that softened the crust in the steeper subduction segment, while the colder lithosphere in the flat slab segment allowed greater crustal thickening and height. From the tectonic point of view the height difference has been attributed to a smaller orogenic contraction in the southern segment. Holocene volcanism appears exclusively at Payun Matru volcano. The associated thermal anomaly may explain the segmentation of the central depression, separating the Cenozoic Los Huarpes basin in the north and the section without coeval sedimentation in the south. The only surface evidence of this segmentation is the E-W, La Carbonilla fault, running both sides of the Payun Matru volcano. Other authors have explained this thermal anomaly as a process of crustal thinning and stretching associated with hot injection from the asthenosphere.
