Descripción: Several species of ecosystem engineers inhabiting coastal environments have been reported structuring different kinds of communities. The magnitude of this influence often depends on the habitat complexity introduced by the engineers. It is commonly accepted that an increase in habitat complexity will result in an increase in diversity and/or abundance in the associated fauna. The rocky salt marshes along the coast of Patagonia are dominated by cordgrasses, mussels, and barnacles forming a mosaic of engineered habitats with different complexity. This system allows us to address the following questions: how different is a macroinvertebrate assemblage when dominated by different ecosystem engineers? And, is there a positive relationship between increasing habitat complexity and the species richness, diversity and total density of the assemblages? To address these questions, we compared the three ecological scenarios with decreasing habitat complexity: cordgrass-mussel, mussel, and barnacle-engineered habitats. We found a total of 22 taxa mostly crustaceans and polychaetes common to all scenarios. The three engineered habitats showed different macroinvertebrate assemblages, mainly due to differences in individual abundances of some taxa. The cryptogenic amphipod Orchestia gammarella was found strictly associated with the cordgrass-mussel habitat. Species richness and diversity were positively related with habitat complexity while total density showed the opposite trend. Our study suggests that species vary their relative distribution and abundances in response to different habitat complexity. Nevertheless, the direction (i. e., neutral, positive or negative) and intensity of the community's response seem to depend on the physiological requirements of the different species and their efficiency to readjust their local spatial distribution in the short term. © 2010 Springer-Verlag and AWI.

Descripción: We analyzed the effect of density-dependent habitat selection and interspecific competition on the use of space by small rodent species in agroecosystems. We studied rodent abundance in cropfields and their borders, weedy margins less disturbed by agricultural activities. We could distinguish two kind of species: those habitat intolerants, Akodon azarae and Oligoryzomys flavescens, which select the borders, and those habitat tolerants, Calomys laucha and Calomys musculinus. The two most abundant species in the community, A. azarae and C. Laucha, differ in their habitat use: while the first has a clear preference for borders, the other is more equally distributed between habitats, although it is more abundant in fields. Both species show a shift towards an increased use of fields via density-dependent habitat selection with increasing abundance at some moments of the year. C. musculinus does not show habitat preference in any season, overlaping with the other species in both habitats. There is an interspecific effect on habitat use between A. azarae and C. laucha : While the first species cause a shift in habitat preference of C. laucha towards fields, this latter species affects the density-dependent habitat use of A. azarae. It is concluded that the pattern of habitat use of the studied species is affected by their differential preferences as well as by intra and interspecific density-dependent processes that change seasonally.

Descripción: An interesting case of 'halogen-bonding-promoted' crystal structure architecture is presented. The two title compounds, C8H8Br2O2 and C8H8I2O2, have almost indistinguishable mol-ecular structures but very different spatial organization, and this is mainly due to differences in the halogen-bonding inter-actions in which the different species present, i.e. Br and I, take part. The dibromo structure exhibits a π-bonded columnar array involving all four independent mol-ecules in the asymmetric unit, with inter-columnar inter-actions governed by C - Br⋯Br - C links and with no C - Br⋯O/N inter-actions present. In the diiodo structure, instead, the C - I⋯O synthon prevails, defining linear chains, in turn inter-linked by C - I⋯I - C inter-actions. © 2008 International Union of Crystallography.

Descripción: Many image processing applications need real time performance, while having restrictions of size, weight and power consumption. These include a wide range of embedded systems from remote sensing applications to mobile phones. FPGA-based solutions are common for these applications, their main drawback being long development time. In this work a co-design methodology for processor-centric embedded systems with hardware acceleration using FPGAs is applied to an image processing method for localization of multiple robots. The goal of the methodology is to achieve a real-time embedded solution using hardware acceleration, but with development time similar to software projects. The final embedded co-designed solution processes 1600×1200 pixel images at a rate of 25 fps, achieving a 12.6× acceleration from the original software solution. This solution runs with a comparable speed as up-to-date PC-based systems, and it is smaller, cheaper and demands less power. © 2012 Springer-Verlag.

Descripción: A compact tabletop plasma focus suitable for hard x-ray imaging applications is presented. The hard x-ray emission was characterized by means of an effective energy analysis, based on images obtained with a single shot. The effective energy of the radiation was estimated to be around 83 keV from radiographic images of metallic pieces. Numerical calculations, based on validated codes, are presented to give a quantitative interpretation of the experimental results. Experimental demonstration of the suitability of this device for introspective imaging of metallic pieces is also given. © 2007 American Institute of Physics.

Descripción: Recent observations of coronal loops in solar active regions show that their heating must be a truly dynamic process. Even though it seems clear that the energy source is the magnetic field that confines the coronal plasma, the details of how it dissipates are still a matter of debate. In this presentation we review the theoretical models of coronal heating, which have been traditionally clasified as DC or AC depending on the electrodynamic response of the loops to the photospheric driving motions. Also, we show results from numerical simulations of the internal dynamics of coronal loops within the framework of the reduced MHD approximation. These simulations indicate that the application of a stationary velocity field at the photospheric boundary leads to a turbulent stationary regime after several photospheric turnover times. Once this turbulent regime is set, both DC and AC stresses dissipate at faster rates as a result of a direct energy cascade. © International Astronomical Union 2011.

Descripción: The aims of this research were: 1) to determine the helminth parasite fauna of seventy two Rattus norvegicus (Berkenhout, 1769) individuals inhabiting poultry farms in the Pampean region of Argentina; 2) to examine the relationship between parasitism and individual characteristics of the hosts, and 3) to analyze the associations among helminths. The study was carried out in twenty-four poultry farms from autumn 2000 to winter 2001. Nematodes were found in the intestine or caecum rectum of the 74 % of rats and cestodes were found along all the intestine of the 28 % of rats. Heterakis spumosa (Travassos 1914) was a central species, whereas Syphacia muris, Yamaguti 1941, Nippostrongylus brasiliensis (Travassos 1914) and Hymenolepis diminuta (Rudolphi 1819) were satellite species. Helminths do not affect rat survival and growth of rats. Rats parasitized with H. spumosa and H. diminuta were larger and older than non-parasitized rats. The intensity of infection with H. spumosa significantly increased with rat age. Rats parasitized with Nippostrongylus brasiliensis and Syphacia muris did not differ in size and age with respect to non-parasitized ones. The mean intensity of infections per host was 33.74, 74.28, higher than 200, and 3.10 for H. spumosa, S. muris, N. brasiliensis and H. diminuta, respectively. The mean intensity of infection with H. spumosa was higher in summer than in the other seasons, while the higher mean intensity of infection with N. brasiliensis and S. muris occurred in autumn. Infections with Heterakis spumosa and Syphacia muris, and with Syphacia muris and Nippostrongylus brasiliensis were independent; while Heterakis spumosa and Nippostrongylus brasiliensis were negatively associated. © 2008 Versita and Springer-Verlag.

Porphyrin biosynthesis from 5-aminoevulinic acid (ALA) was investigated using the technique of tissue explant cultures, in both human breast cancer and its original normal tissue. 2. 2. The activity of ALA-dehydratase, porphobilinogenase and uroporphyrinogen decarboxylase was directly determined in both tumor and normal mammary tissues. 3. 3. Porphyrin synthesis capacity of human breast carcinoma was 20-fold enhanced, as compared with normal tissue, at least between the stages of porphobilinogen and coproporphyrinogen formation. 4. 4. The activity of the three enzymes examined was always lower in normal tissue than in tumoral tissue. 5. 5. Basal levels and allyl-isopropylacetamide (AIA) or veronal induced levels of δ-aminolevulinate synthetase (ALA-S), cytochrome P-450 (cyt P-450) and cytochrome oxidase were determined in tumor (T) and liver of both normal mice (NM) and T bearing mice (TBM). 2. 2. Basal levels of ALA-S were nearly the same in either source. The amount of cyt P-450 was lower in TBM liver than in NM liver, and no detectable in T. While the basal activity of cytochrome oxidase in TBM liver and T were higher than those of NM liver. 3. 3. In AIA intoxicated animals there was a lower induction of ALA-S in liver of TBM than in NM liver. There was no induction in T ALA-S. The loss of cyt P-450 was less in TBM liver when compared with NM liver. 4. 4. The induction level of cyt P-450 after veronal administration was nearly the same in liver of both TBM and NM. 5. 5. We conclude that lower induction of liver ALA-S activity in TBM liver is due to correspondingly lower drug metabolism ability of TBM liver. Otherwise our results suggest that the control mechanism operating in T and probably in its original tissue are different from those described for normal liver. © 1990.

The activity of Suc.CoA-S and ALA-S were twice higher in symbiote-containing than in aposymbiotic C. deanei. Aposymbiotic cells had a higher activity of DOVA-T than symbiote-containing cells. 5. 5. Descripción: Although the negative regulation of gene expression by insulin has been widely studied, the transcription factors responsible for the insulin effect are still unknown. The purpose of this work was to explore the molecular mechanisms involved in the insulin repression of the 5-aminolevulinate synthase (ALAS) gene. Deletion analysis of the 5′-regulatory region allowed us to identify an insulin-responsive region located at -459 to -354 bp. This fragment contains a highly homologous insulin-responsive (IRE) sequence. By transient transfection assays, we determined that hepatic nuclear factor 3 (HNF3) and nuclear factor 1 (NF1) are necessary for an appropriate expression of the ALAS gene. Insulin overrides the HNF3β or HNF3β plus NF1-mediated stimulation of ALAS transcriptional activity. Electrophoretic mobility shift assay and Southwestern blotting indicate that HNF3 binds to the ALAS promoter. Mutational analysis of this region revealed that IRE disruption abrogates insulin action, whereas mutation of the HNF3 element maintains hormone responsiveness. This dissociation between HNF3 binding and insulin action suggests that HNF3β is not the sole physiologic mediator of insulin-induced transcriptional repression. Furthermore, Southwestern blotting assay shows that at least two polypeptides other than HNF3β can bind to ALAS promoter and that this binding is dependent on the integrity of the IRE. We propose a model in which insulin exerts its negative effect through the disturbance of HNF3β binding or transactivation potential, probably due to specific phosphorylation of this transcription factor by Akt. In this regard, results obtained from transfection experiments using kinase inhibitors support this hypothesis. Due to this event, NF1 would lose accessibility to the promoter. The posttranslational modification of HNF3 would allow the binding of a protein complex that recognizes the core IRE. These results provide a potential mechanism for the insulin-mediated repression of IRE-containing promoters.

Descripción: Heregulin (HRG) and type I receptor tyrosine kinase (RTK) expression was investigated in the highly invasive and metastatic LM3 cell line, our previously described model of metastasis for mammary cancer (Bal de Kier Joffe et al. [1986] Invasion Metastasis 6:302-12; Urtreger et al. [1997] Int J Oncol 11:489-96). Although LM3 cells do not express HRG, they exhibit high levels of ErbB-2 and ErbB-3 as well as moderate expression of ErbB-4. Addition of exogenous HRGβ1 resulted in inhibition of both proliferation and migration of LM3 cells. HRGβ1 was also able to decrease the activity of urokinase-type plasminogen activator (uPA) and matrix metalloproteinase 9 (MMP-9), 2 key enzymes in the invasion and metastatic cascade. HRGβ1 treatment of LM3 cells induced tyrosine phosphorylation of ErbB-2, ErbB-3 and ErbB-4 as well as the formation of ErbB-2/ErbB-3 and ErbB-2/ErbB-4 heterodimers. Assessment of the signaling pathways involved in HRGβ1 action indicated that the addition of HRGβ1 to LM3 cells resulted in activation of phosphatidylinositol 3-kinase (PI-3K) and in strong induction of the association of the p85 subunit of PI-3K with ErbB-3. HRGβ1 also caused the rapid activation of ERKI/ERK2 and Stat3 and Stat5 (signal transducers and activators of transcription [STAT]). This is the first demonstration of the ability of HRGβ1 to activate STATs in mammary tumor cells. Blockage of PI-3K activity with its chemical inhibitor wortmannin, or of MEKI/ERKs activity with PD98059, resulted in suppression of the ability of HRGβ1 to inhibit LM3 cell growth. Notwithstanding the suppression of these 2 signaling pathways, HRGβ1 still proved capable of inhibiting uPA activity. Therefore, our results provide evidence that signaling pathways involved in HRGβ1-induced proliferation appear to be distinct from those involved in HRGβ1 regulation of uPA, a protease that plays a pivotal role in invasion and metastasis. © 2002 Wiley-Liss, Inc.

Descripción: Populations may adapt in response to selection pressures imposed by global environmental change. In marine zooplankton, measurements of the heritability of key life history characters, and thus the potential for evolution, are still rare. Here we demonstrate the feasibility of conducting controlled mating experiments with the dioecious appendicularian Oikopleura dioica to explore the narrow-sense heritability and genetic correlation among morphological and life history traits. At our standard laboratory conditions (15 ± 1°C, 100 μg C l-1), mature females were larger (1.213 ± 0.19 mm, mean ± SD) and lived longer (8.5 ± 2.18 d) than did males (1.115 ± 0.15 mm, 7.6 ± 2.07 d). The heritability (±SE) of morphological characters was low (trunk size, 0.37 ± 0.25; house size, 0.39 ± 0.23) to moderate (tail length, 0.50 ± 0.31). In contrast, an important life history trait, lifespan, showed high heritability (0.89 ± 0.47) and may therefore respond rapidly to selection pressure, either in the laboratory or in the wild. © Inter-Research 2011.

Descripción: We have developed multicellular spheroids (MCS) established from LM05e and LM3 spontaneous Balb/c-murine mammary adenocarcinoma and B16 C57-murine melanoma derived cell lines as an in vitro model to study the efficacy of the herpes simplex virus thymidine kinase/ganciclovir (HSVtk/GCV) suicide system. We demonstrated for the first time that HSVtk-expressing cells assembled as MCS manifested a GCV resistance phenotype compared to the same cells grown as sparse monolayers. HSVtk-expressing LM05e, LM3 and B16 spheroids were 16-, three- and nine-fold less sensitive to GCV than their respective monolayers, even though they could express transgenes 10-, eight- and five-fold more efficiently. Mixed populations of HSVtk- and their respective βgal-expressing cells displayed a cell-type specific bystander effect that was higher in monolayers than in MCS. However, HSVtk-expressing cells in two- or three-dimensional cultures were always significantly more sensitive to GCV than the βgal-expressing counterparts, supporting the feasibility of this suicide approach in vivo. We present evidence showing that HSVtk-expressing tumor cells, when transferred from monolayers to MCS, displayed: (i) lower GCV cytotoxic activity and bystander effect; (ii) higher and efficient expression of genes transferred as lipoplexes; (iii) lower cell proliferation rates; and (iv) changes in intracellular Bax/Bcl-xL rheostat of mitochondria-mediated apoptosis.

Descripción: The plant aquaporin plasma membrane intrinsic proteins (PIP) subfamily represents one of the main gateways for water exchange at the plasma membrane (PM). A fraction of this subfamily, known as PIP1, does not reach the PM unless they are coexpressed with a PIP2 aquaporin. Although ubiquitous and abundantly expressed, the role and properties of PIP1 aquaporins have therefore remained masked. Here, we unravel how FaPIP1;1, a fruit-specific PIP1 aquaporin from Fragaria x ananassa, contributes to the modulation of membrane water permeability (Pf) and pH aquaporin regulation. Our approach was to combine an experimental and mathematical model design to test its activity without affecting its trafficking dynamics. We demonstrate that FaPIP1;1 has a high water channel activity when coexpressed as well as how PIP1-PIP2 affects gating sensitivity in terms of cytosolic acidification. PIP1-PIP2 random heterotetramerization not only allows FaPIP1;1 to arrive at the PMbut also produces an enhancement of FaPIP2;1 activity. In this context, we propose that FaPIP1;1 is a key participant in the regulation of water movement across the membranes of cells expressing both aquaporins.

Descripción: A list of 32 species of Heteroptera captured with pitfall traps at INTA (Instituto Nacional de Tecnología Agropecuaria) Delta del Paraná (Partido de Campana, Buenos Aires) is presented in this paper. Two species are mentioned as a first record for the province: Fulvius bisbistillatus (Stål) (Miridae) and Pselliopus ornaticeps (Stål) (Reduviidae). Also an analysis of Spearman rank correlation ® and correspondence analysis is made, to assess the spatial and temporal variation of Heteroptera communities of different selected land uses.

Descripción: Female birds that engage in extrapair mating may choose extrapair mates that are genetically compatible, increasing their fitness through genetic benefits, such as increased heterozygosity, to their offspring; or choose mates that are heterozygous at one or more loci. Here, we describe the extrapair mating system, explore the fitness benefits of extrapair mating and test the heterozygosity hypothesis in White-rumped Swallows (Tachycineta leucorrhoa) breeding in Argentina using a panel of microsatellite loci. Extrapair offspring accounted for 56% of the nestlings and 77% of the broods in our population. Within broods, 1-4 males fathered extrapair offspring, and in 29% of nests, all offspring were from extrapair sires. We found that broods with extrapair offspring fledged overall more young than broods with no extrapair offspring but that the young that died were more heterozygous than the ones that fledged. Although extrapair offspring had a higher probability of surviving than within-pair offspring, these 2 groups did not differ in their level of heterozygosity. Neither the heterozygosity of the social mate nor the genetic similarity of the social pair predicted the presence of extrapair young. Instead, females chose social mates that were significantly less genetically similar to them. Our results do not support the heterozygosity hypothesis and contradict 2 of its main predictions. © 2011 The Author. Descripción: The lipopeptidophosphoglycan from Trypanosoma cruzi is a glycosylated inositol‐phosphoceramide isolated from epimastigotes at the stationary phase of growth (4–5 days). We have now purified two similar glycoinositolphospholipids (glycoinositolphospholipid A and glycoinositolphospholipid B) from epimastigotes after the second day of culture growth. [3H]Palmitic acid was incorporated into 1‐O‐hexadecyl‐2‐O‐palmitoylglycerol in glycoinositolphospholipid A and into ceramide in glycoinositolphospholipid B. The lipids were released by incubation with glycosylphosphatidylinositol‐specific phospholipase C from Bacillus thuringiensis or by chemical methods. After alkaline hydrolysis, the lipids were analysed by GLC/MS. In glycoinositolphospholipid A the resulting lipids corresponded to 1‐O‐hexadecylglycerol and palmitic acid. The ceramide components in glycoinositolphospholipid B are sphinganine, palmitic acid and lignoceric acid. The oligosaccharides could be degraded by nitrous acid and further enzymic treatment showed that the two glycoinositolphospholipids isolated from T. cruzi share the common core structure of the glycosylphosphatidylinositol membrane anchors. The microheterogeneity was determined, as well as the substitution by galactose, and was mainly in the furanose configuration as was previously described for lipopeptidophosphoglycan. However, methylation analysis indicated that 20% of the galactose is in the pyranose from. Both glycoinositolphospholipids mainly differ in the lipid moiety. Copyright © 1993, Wiley Blackwell. Descripción: We present updated predictions for the total cross section for Higgs boson production by gluon-gluon fusion in hadron collisions. Our calculation includes the most advanced theoretical information available at present for this observable: soft-gluon resummation up to next-to-next-to-leading logarithmic accuracy, the exact treatment of the bottom-quark contribution up to next-to-leading order, and two-loop electroweak effects. We adopt the most recent parametrization of parton distribution functions at next-to-next-to-leading order, and we evaluate the corresponding uncertainties. In comparison with our previous central predictions, at the Tevatron the difference ranges from +9% for mH = 115   GeV to -9% for mH = 200   GeV. At the LHC the cross section is instead significantly increased. The effect goes from +30% for mH = 115   GeV to +9% for mH = 300   GeV, and is mostly due to the new parton distribution functions. We also provide new predictions for the LHC at sqrt(s) = 10   TeV. © 2009 Elsevier B.V.

Descripción: The geological observations made by Darwin in 1835 during his crossing of the Andes from Santiago to Mendoza via the Piuquenes Pass and Cordón del Portillo are compared with the present geological knowledge of the Cordillera Principal and Cordillera Frontal at 33°-34°S. The analysis of the complex stratigraphy of the Cordillera Principal, the imbricated structure of the Aconcagua fold and thrust belt, as well as the stratigraphy and structure of the inter mountain foreland Tunuyán Basin, allows to assess the pioneer observations of Darwin. He recognized the old metamorphic basement and the granitoids and volcanic sequences of late Paleozoic to Triassic age of the Cordillera Frontal, established the Cretaceous age of the marine successions cropping out along the eastern Cordillera Principal and studied the conglomeratic deposits associated with the uplift of the Cordillera in the Alto Tunuyán Basin. Based on the study of clast provenance of the synorogenic deposits of the Alto Tunuyán Basin, Darwin recognized that the Cordillera Frontal was uplifted later than the Cordillera Principal. The present knowledge of this sector of the Andean Cordillera confirms his pioneer observations and show that Darwin was one of the first scientists ever in realizing that in an orogenic system the sequence of uplift and deformation proceeds from hinterland towards foreland, according to a process that is exceptionally well-illustrated along the Piuquenes-Cordón del Portillo transect.

Descripción: Mucins are highly O-glycosylated molecules which in mammalian cells accomplish essential functions, like cytoprotection and cell-cell interactions. In the protozoan parasite Trypanosoma cruzi, mucin-related glycoproteins have been shown to play a relevant role in the interaction with and invasion of host cells. We have previously reported a family of mucin- like genes in T. cruzi whose overall structure resembled that of mammalian mucin genes. We have now analyzed the relationship between these genes and mucin proteins. A monoclonal antibody specific for a mucin sugar epitope and a polyclonal serum directed to peptide epitopes in a MUC gene-encoded recombinant protein, detected identical bands in three out of seven strains of T. cruzi. Immunoprecipitation experiments confirmed these results. When expressed in eukaryotic cells, the MUC gene product is post-translationally modified, most likely, through extensive O-glycosylation. Gene sequencing showed that the central domains encoding the repeated sequences with the consensus T 8KP 2, varies in number from 1 to 10, and the number of Thr residues in each repeat could be 7, 8, or 10. A run of 16 to 18 Thr residues was present in some, but not all, MUC gene-derived sequences. Direct compositional analysis of mucin core proteins showed that Thr residues are much more frequent than Ser residues. The same fact occurs in MUC gene- derived protein sequences. Molecular mass determinations of the 35-kDa glycoproteins further extend the heterogeneity of the family to the natural mucin molecules. Difficulties in assigning each of the several MUC genes identified to a mucin product arise from the high diversity and partial sequence conservation of the members of this family.

Descripción: Aims.This paper reports molecular observations towards the bright eastern knot (BEK) in the SNR Puppis A, a feature where radio and X-ray studies suggest that the shock front is interacting with a dense molecular clump.Methods.We performed high-resolution millimetric observations towards the BEK of Puppis A using the SEST telescope in the CO -0 and 2-1 lines (beams of 45 and 23, respectively). More extended, lower angular resolution CO -0 observations taken from NANTEN archival data were also analyzed to obtain a complete picture.Results. In the velocity range near 16 km s-1, which is the Puppis A systemic velocity, our study revealed two important properties: (i) no dense molecular gas is detected immediately adjacent to the eastern border of the BEK and (ii) the molecular clump detected very close to the radiocontinuum maximum is probably located in the foreground along the line of sight and has not yet been reached by the SNR shock front. We propose two possible scenarios for explaining the absence of molecular emission eastwards of the BEK border of Puppis A. Either the shock front has completely engulfed and destroyed a molecular clump or the shock front is interacting with part of a larger cloud, and we do not detect CO emission immediately beyond it because the molecules have been dissociated by photodissociation and by reactions with photoionized material due to the radiative precursor. © 2008 ESO.

Descripción: Aims.We investigate the morphology at low radio frequencies of the supernova remnant (SNR) IC 443 in detail and accurately establish its radio continuum spectral properties. Methods. We used the VLA in multiple configurations to produce high-resolution radio images of IC 443 at 74 and 330 MHz. From these data we produced the first sensitive, spatially resolved spectral analysis of the radio emission at long wavelengths. The changes with position in the radio spectral index were correlated with data in near infrared (NIR) from 2MASS, in gamma-rays from VERITAS, and with the molecular 12CO (J = 1-0) line emission. Results. The new image at 74 MHz has HPBW = 35′′ and rms = 30 mJy beam-1 and at 330 MHz HPBW = 17′′and rms = 1.7 mJy beam-1. The integrated flux densities for the whole SNR are S SNR 74MHz = 470 ± 51 Jy and S SNR 330MHz = 248 ± 15 Jy. Improved estimates of the integrated spectrum were derived taking a turnover into account to fit the lowest frequency measurements in the literature. Combining our measurements with existing data, we derive an integrated spectral index α 10700MHz 10MHz =-0.39 ± 0.01 with a free-free continuum optical depth at 330 MHz 330 ∼ 7 × 10 -4 (τ10 = 1.07); all measurements above ∼10 MHz are equally consistent with a power law spectrum. For the pulsar wind nebula associated with the compact source CXOU J061705.3+222127, we calculated S PWN 330MHz = 0.23±0.05 Jy, SPWN 1420MHz = 0.20±0.04 Jy, and α8460MHz 330MHz ∼ 0.0. Substantial variations are observed in spectral index between 74 and 330 MHz across IC 443. The flattest spectral components (-0.25 = a =-0.05) coincide with the brightest parts of the SNR along the eastern border, with an impressive agreement with ionic lines as observed in the 2MASS J and H bands. The diffuse interior of IC 443 has a spectrum steeper than found anywhere in the SNR (-0.85 = a =-0.6), while the southern ridge again has a flatter spectrum (a ∼-0.4). With the available statistics the VERITAS .-ray emission strikingly matches the CO distribution, but no clear evidence is found for a morphological correlation between the TeV distribution and radio emission. Conclusions. The excellent correspondence between the eastern radio flattest spectrum region and NIR ionic lines strongly suggests that the passage of a fast, dissociating J-type shock across the interacting molecular cloud dissociated the molecules and ionized the gas.We therefore conclude that thermal absorption at 74 MHz (τ74 up to ∼0.3) is responsible for the localized spectral index flattening observed along the eastern border of IC 443. Towards the interior of IC 443, the spectrum is consistent with those expected from linear diffusive shock acceleration, while the flatter spectrum in the southern ridge is a consequence of the strong shock/molecular cloud interaction. © 2011 ESO.

Descripción: p8 is a nuclear DNA-binding protein, which was identified because its expression is strongly activated in response to several stresses. Biochemical and biophysical studies revealed that despite a weak sequence homology p8 is an HMG-I/Y-like protein, suggesting that p8 may be involved in transcription regulation. Results reported here strongly support this hypothesis. Using a pull-down approach, we found that p8 interacts with the general co-activator p300. We also found that, similar to the HMG proteins, p300 was able to acetylate recombinant p8 in vitro, although the significance of such modification remains to be determined. Then a screening by the two-hybrid system, using p8 as bait, allowed us to identify the Pax2 trans-activation domain-interacting protein (PTIP) as another partner of p8. Transient transfection studies revealed that PTIP is a strong inhibitor of the trans-activation activities of Pax2A and Pax2B on the glucagon gene promoter, which was chosen as a model because it is a target of the Pax2A and Pax2B transcription factors. This effect is completely abolished by co-transfection of p8 in glucagon-producing InRIG9 cells, indicating that p8 binding to PTIP prevents inhibition of the glucagon gene promoter. This was not observed in NIH3T3 fibroblasts that do not express glucagon. Finally, expression of p8 enhances the effect of p300 on Pax2A and Pax2B trans-activation of the glucagon gene promoter. These observations suggest that in glucagon-producing cells p8 is a positive cofactor of the activation of the glucagon gene promoter by Pax2A and Pax2B, both by recruiting the p300 cofactor to increase the Pax2A and Pax2B activities and by binding the Pax2-interacting protein PTIP to suppress its inhibition.

Descripción: Background: Chronic injury deregulates cellular homeostasis and induces a number of alterations leading to disruption of cellular processes such as cell cycle checkpoints and apoptosis, driving to carcinogenesis. The stress protein heme oxygenase-1 (HO-1) catalyzes heme degradation producing biliverdin, iron and CO. Induction of HO-1 has been suggested to be essential for a controlled cell growth. The aim of this work was to analyze the in vivo homeostatic response (HR) triggered by the withdrawal of a potent carcinogen, p-dimethylaminoazobenzene (DAB), after preneoplastic lesions were observed. We analyzed HO-1 cellular localization and the expression of HO-1, Bcl-2 and cell cycle related proteins under these conditions comparing them to hepatocellular carcinoma (HC). Methods: The intoxication protocol was designed based on previous studies demonstrating that preneoplastic lesions were evident after 89 days of chemical carcinogen administration. Male CF1 mice (n = 18) were used. HR group received DAB (0.5 % w/w) in the diet for 78 days followed by 11 days of carcinogen deprivation. The HC group received the carcinogen and control animals the standard diet during 89 days. The expression of cell cycle related proteins, of Bcl-2 and of HO-1 were analyzed by western blot. The cellular localization and expression of HO-1 were detected by immnunohistochemistry. Results: Increased expression of cyclin E/CDK2 was observed in HR, thus implicating cyclin E/CDK2 in the liver regenerative process. p21cip1/waf1 and Bcl-2 induction in HC was restituted to basal levels in HR. A similar response profile was found for HO-1 expression levels, showing a lower oxidative status in the carcinogen-deprived liver. The immunohistochemical studies revealed the presence of macrophages surrounding foci of necrosis and nodular lesions in HR indicative of an inflammatory response. Furthermore, regenerative cells displayed changes in type, size and intensity of HO-1 immunostaining. Conclusion: These results demonstrate that the regenera tive capacity of the liver is still observed in the pre-neoplastic tissue after carcinogen withdrawal suggesting that reversible mechanism/s to compensate necrosis and to restitute homeostasis are involved. © 2006 Castronuovo et al; licensee BioMed Central Ltd.

Descripción: Recent experimental and theoretical methods allowed the efficient investigation of highly excited rovibrational states of molecular systems. At these levels of excitation the correspondence principle holds, and then classical mechanics can provide intuitive views of the involved processes. In this respect, we have recently shown that for completely hyperbolic systems, homoclinic motions, which are known to organize the classical chaotic region in Hamiltonian systems, imprint a clear signature in the corresponding highly excited quantum spectra. Descripción: In a previous paper, we have modeled the coupling between corona and chromosphere and derived a non-linear set of equations, where the global stability properties of the coronal plasma can be studied. The linear stability analysis indicates that the static equilibrium is stable unless the heating rate falls below a certain critical value. In the present paper, we study the nonlinear evolution of our equations both analytically and numerically. Applying a perturbative technique around the critical point, we find that a subcritical Hopf bifurcation takes place. The numerical integration of the equations agrees satisfactorily with the analytical results when they are compared close to the bifurcation. The nonthermal Doppler widths of EUV lines forming in the transition region can be explained by the existence of relatively low amplitude limit cycles.

Descripción: We study the coupling between the hot plasma confined in a coronal loop and the much colder chromospheric plasma at the footpoints. Considering the coronal heating rate as a control parameter, we find that the static equilibrium becomes unstable for heating rates below a critical value, giving rise to the appearance of a stable limit cycle. Starting from the hydrodynamic equations, we derive a model which generalizes the analysis of Kuin and Martens and consistently takes into account the condensation-evaporation process. In this paper, we linearize our equations in order to find the bifurcation point where the stability of the static equilibrium is lost. We also show that this model can provide a natural explanation for the excess widths of EUV spectral lines formed in the transition region. Moreover, we can predict the observed reduction in the broadening of these lines when they form in certain active regions, like quiescent prominences or sunspots.

Descripción: Continuity and evolution in time of the horizontal stress direction in the Neuquen Basin area, derived from the analysis of recent borehole data and orientation of volcanic dykes measured in outcrop in the Cara Cura and Reyes Ranges is presented. The bitumen dykes along the Neuquén Basin were formed during Paleocene - Eocene in a context where the maximum stress was horizontal and had a NE. Within the analyzed volcanic dykes two major groups have been recognized, group i (NE orientation) and group ii (NNE orientation) and a secondary group iii (NW orientation). The age of these dykes is still not well established, but they were related to a Late Oligocene - Miocene magmatic event. Both for the volcanic and bitumen dykes a type i group (ENE-NE orientation) has been recognized, evidencing the maximum stress direction at the time they were formed. Beyond the uncertainties in the volcanic and bitumen dykes age, it is interpreted that during great part of Tertiary, the orientation of the horizontal maximum stress was NE. However, the breakout data shows that the actual horizontal maximum stress is a little different with a ENE mean direction. This change in the direction of stress is consistent with the change in the subduction vector produced between the Eocene and today.

Descripción: Trypanosoma cruzi, etiological agent of Chagas' disease, has a complex life cycle which involves the invasion of mammalian host cells, differentiation and intracellular replication. Here we report the first insights into the biological role of a poly(ADP-ribose) glycohydrolase in a trypanosomatid (TcPARG). In silico analysis of the TcPARG gene pointed out the conservation of key residues involved in the catalytic process and, by Western blot, we demonstrated that it is expressed in a life stage-dependant manner. Indirect immunofluorescence assays and electron microscopy using an anti-TcPARG antibody showed that this enzyme is localized in the nucleus independently of the presence of DNA damage or cell cycle stage. The addition of poly(ADP-ribose) glycohydrolase inhibitors ADP-HPD (adenosine diphosphate (hydroxymethyl) pyrrolidinediol) or DEA (6,9-diamino-2-ethoxyacridine lactate monohydrate) to the culture media, both at a 1 μM concentration, reduced in vitro epimastigote growth by 35% and 37% respectively, when compared to control cultures. We also showed that ADP-HPD 1 μM can lead to an alteration in the progression of the cell cycle in hydroxyurea synchronized cultures of T. cruzi epimastigotes. Outstandingly, here we demonstrate that the lack of poly(ADP-ribose) glycohydrolase activity in Vero and A549 host cells, achieved by chemical inhibition or iRNA, produces the reduction of the percentage of infected cells as well as the number of amastigotes per cell and trypomastigotes released, leading to a nearly complete abrogation of the infection process. We conclude that both, T. cruzi and the host, poly(ADP-ribose) glycohydrolase activities are important players in the life cycle of Trypanosoma cruzi, emerging as a promising therapeutic target for the treatment of Chagas' disease. © 2013 Vilchez Larrea et al.

Descripción: Small outcrops of biotitic-amphibolic schists and amphibolites found in two localities suggest that the Cushamen Formation is host rock of the Central Patagonian Batholith in Gastre. The characteristics of these rocks were investigated using structural, petrographic, magnetic fabric, and garnet chemical-zonation studies. Garnet is observed as a late product of contact metamorphism. Structural data from the studied area and from other localities in the region are in conflict with models that invoke the occurrence of uniform, large-scale dextral displacements along the Gastre lineament in Mesosoic times. Rather, the data suggest that the Gastre granitoids record heterogeneous low-temperature deformation of likely Late Triassic age. This low-temperature deformation appears as small, localized outcrops, and we suggest that it is related to the accommodation of the successive magma batches that built the Central Patagonian Batholith.

Descripción: Variable retention harvesting (VRH), in which trees are removed at variable intensity and spatial configuration across the landscape, retains greater forest structural heterogeneity than traditional clear-cut harvesting and is being recommended as an alternative for sustainable management of the boreal forest. Little is known about its effects on forest fauna; thus, we studied the influence of one type of VRH (harvesting with advanced regeneration [HARP]) on the Syrphidae (Diptera) community in northern Ontario forests of peatland black spruce (Picea mariana). We examined the effects of varying structural retention (from unharvested through partial retention to clear-cut) on syrphid species richness and abundance, and abundance of functional assemblages. Greater species richness and population abundances were found generally in harvested than in unharvested forests. Overall species richness and the abundance of four species (Platycheirus rosarum, Toxomerus marginatum, Xylota annulifera, and X. tuberculata) and larval predators were all higher in both clear-cut sites and those with structural retention than in unharvested sites. Similarly, overall species richness and the abundance of nine species were higher in clear-cut than in unharvested sites. Species responses are discussed in an ecological context. Differences among the levels of forest retention harvesting were relatively minor compared with those of the clear-cut and unharvested area, suggesting that local habitat characteristics may play a more important role in determining the syrphid community than the landscape configuration. However, a landscape level effect was evident, suggesting that syrphids may be useful in reflecting changes in stand structure at the landscape scale. © 2007 Entomological Society of America.

Descripción: Apis mellifera bees perform dances to communicate the presence of desirable nectar sources. The regulation of these dances does not depend exclusively on properties of the nectar sources, but also upon certain stimuli derived from the foraging status of the colony as a whole; i.e. bees exploiting a source of constant profitability are more likely to dance when the colony's nectar intake rate is low. Based on these stimuli, individual bees tune their dances according to their colony's nectar influx without visiting alternative nectar sources. Division of labour, in addition, is a common feature in honeybees. Upon returning to the nest, successful foragers transfer the content of their crops to food-receivers by means of a common behaviour in social insects called trophallaxis, i.e. the transfer of liquid food by mouth. Martin Lindauer stated that a returned forager may sense the foraging status of its colony on the basis of the food transfer process by computing how quickly and eagerly the food-receivers unload its crop. This study focuses on the forager's experience during the food transfer process, its variability based on the colony's nectar influx, and the separate effects that the 'ease' and the 'eagerness' of the food-unloading have on the tuning of recruitment dances. Results indicate that foragers can rapidly sense variations in the colony's nectar influx, even when they experience no variation in the time interval between their return to the hive and the beginning of the food transfer. To accomplish this task they appear to use stimuli derived from the number of food-receivers, which enable them, in turn, to set their dance thresholds in relation to the nectar influx of their colony. The relevance of these findings is discussed in the context of communication and successful foraging.

Descripción: The tumor suppressor protein p53 is known to be transported to the nucleus along microtubular tracks by cytoplasmic dynein. However, the connection between p53 and the dynein motor protein complex has not been established. Here, we show that hsp90·binding immunophilins link p53·hsp90 complexes to dynein and that prevention of that linkage in vivo inhibits the nuclear movement of p53. First, we show that p53·hsp90 heterocomplexes from DLD-1 human colon cancer cells contain an immunophilin (FKBP52, CyP-40, or PP5) as well as dynein. p53·hsp90·immunophilin·dynein complexes can be formed by incubating immunopurified p53 with rabbit reticulocyte lysate, and we show by peptide competition that the immunophilins link via their tetratricopeptide repeat domains to p53-bound hsp90 and by means of their PPIase domains to the dynein complex. The linkage of immunophilins to the dynein motor is indirect by means of the dynamitin component of the dynein-associated dynactin complex, and we show that purified FKBP52 binds directly by means of its PPIase domain to purified dynamitin. By using a temperature-sensitive mutant of p53 where cytoplasmic-nuclear movement occurs by shift to permissive temperature, we show that p53 movement is impeded when p53 binding to hsp90 is inhibited by the hsp90 inhibitor radicicol. Also, nuclear movement of p53 is inhibited when immunophilin binding to dynein is competed for by expression of a PPIase domain fragment in the same manner as when dynein linkage to cargo is dissociated by expression of dynamitin. This is the first demonstration of the linkage between an hsp90-chaperoned transcription factor and the system for its retrograde movement to the nucleus both in vitro and in vivo.

Descripción: In this study, we demonstrate that the subcellular localization of the mineralocorticoid receptor (MR) is regulated by tetratricopeptide domain (TPR) proteins. The high-molecular-weight immunophilin (IMM) FKBP52 links the MR-hsp90 complex to dynein/dynactin motors favoring the cytoplasmic transport of MR to the nucleus. Replacement of this hsp90-binding IMM by FKBP51 or the TPR peptide favored the cytoplasmic localization of MR. The complete movement machinery, including dynein and tubulin, could be recovered from paclitaxel/GTP-stabilized cytosol and was fully reassembled on stripped MR immune pellets. The whole MR-hsp90-based heterocomplex was transiently recovered in the soluble fraction of the nucleus after 10 min of incubation with aldosterone. Moreover, cross-linked MR-hsp90 heterocomplexes accumulated in the nucleus in a hormone-dependent manner, demonstrating that the heterocomplex can pass undissociated through the nuclear pore. On the other hand, a peptide that comprises the DNA-binding domain of MR impaired the nuclear export of MR, suggesting the involvement of this domain in the process. This study represents the first report describing the entire molecular system that commands MR nucleocytoplasmic trafficking and proposes that the MR-hsp90-TPR protein heterocomplex is dissociated in the nucleus rather than in the cytoplasm. Descripción: Two populations of human natural killer (NK) cells can be identified in peripheral blood. The majority are CD3-CD56dim cells while the minority exhibits a CD3-CD56bright phenotype. In vitro evidence indicates that CD56bright cells are precursors of CD56dim cells, but in vivo evidence is lacking. Here, we studied NK cells from a patient that suffered from a melanoma and opportunistic fungal infection during childhood. The patient exhibited a stable phenotype characterized by a reduction in the frequency of peripheral blood CD3-CD56dim NK cells, accompanied by an overt increase in the frequency and absolute number of CD3-CD56bright cells. These NK cells exhibited similar expression of perforin, CD57 and CD158, the major activating receptors CD16, NKp46, NKG2D, DNAM-1, and 2B4, as well as the inhibitory receptor CD94/NKG2A, on both CD56bright and CD56dim NK cells as healthy controls. Also, both NK cell subpopulations produced IFN-γ upon stimulation with cytokines, and CD3-CD56dim NK cells degranulated in response to cytokines or K562 cells. However, upon stimulation with cytokines, a substantial fraction of CD56dim cells failed to up-regulate CD57 and CD158, showed a reduction in the percentage of CD16+ cells, and CD56bright cells did not down-regulate CD62L, suggesting that CD56dim cells could not acquire a terminally differentiated phenotype and that CD56bright cells exhibit a maturation defect that might result in a potential altered migration pattern. These observations, support the notion that NK cells of this patient display a maturation/activation defect that precludes the generation of mature NK cells at a normal rate accompanied by CD56dim NK cells that cannot completely acquire a terminally differentiated phenotype. Thus, our results provide evidence that support the concept that in vivo CD56bright NK cells differentiate into CD56dim NK cells, and contribute to further understand human NK cell ontogeny. © 2012 Domaica et al.

Descripción: We have studied the biochemical features, the conformational preferences in solution, and the DNA binding properties of human p8 (hp8), a nucleoprotein whose expression is affected during acute pancreatitis. Biochemical studies show that hp8 has properties of the high mobility group proteins, HMG-I/Y. Structural studies have been carried out by using circular dichroism (near- and far-ultraviolet), Fourier transform infrared, and NMR spectroscopies. All the biophysical probes indicate that hp8 is monomeric (up to 1 mM concentration) and partially unfolded in solution. The protein seems to bind DNA weakly, as shown by electrophoretic gel shift studies. On the other hand, hp8 is a substrate for protein kinase A (PKA). The phosphorylated hp8 (PKAhp8) has a higher content of secondary structure than the nonphosphorylated protein, as concluded by Fourier transform infrared studies. PKAhp8 binds DNA strongly, as shown by the changes in circular dichroism spectra, and gel shift analysis. Thus, although there is not a high sequence homology with HMG-I/Y proteins, hp8 can be considered as a HMG-I/Y-like protein.

Descripción: Humic substances (HS) were isolated from the sediments of Lobos Pond (Argentina) using mild conditions to preserve their native structure. The HS (humic and fulvic acids) were characterized by means of elemental analysis and FTIR spectroscopy. Also a by-product obtained during fulvic acids (FA) fractionation (an amorphous white solid residue) was analyzed. Results revealed possible interactions between FA and inorganic-organic substances that may have implications referring to bioavailability. Other limnological implications, such as autochtonous origin of HS linked with the hydrology, and change of pH during stormy weather that affects HS interactions, are discussed.

Descripción: Several studies indicate that hyaluronan oligosaccharides (oHA) are able to modulate growth and cell survival in solid tumors; however, no studies have been undertaken to analyze the effect of oHA on T-lymphoid disorders. In this work we showed that oHA were able to induce apoptosis in lymphoma cell lines. Since PI3-K/Akt and nuclear factor-κB (NF-κB) are major factors involved in cell survival and anti-apoptotic pathways in lymphoma cells, we hypothesized that oHA could induce apoptosis through inhibition of these pathways. oHA were identified by a method which allows characterization of length using a high pH anion exchange chromatography with pulse amperometric detection (HPAEC-PAD). oHA inhibited PIP3 production (principal product of PI3-K activity) and reduced Akt phosphorylation levels, similarly to the specific inhibitor wortmannin. However, treatment with either oHA or wortmannin failed to inhibit constitutive NF-κB activity and modulate IκBα protein levels, suggesting that PI3-K and NF-κB signaling pathways are not related in the cell lines used. Cell behavior differed using native hyaluronan (HA), which induced PIP3 production, Akt phosphorylation, and NF-κB activation, although not related with cell survival since treatment with native HA showed no effect on apoptosis. Our results suggest that oHA induce apoptosis by suppression of PI3-K/Akt cell survival pathway without involving NF-κB activation, through a mechanism that differs from the one mediated by native HA. © 2006 Oxford University Press.

Descripción: We study systems of three bivariate polynomials whose Newton polygons are scaled copies of a single polygon. Our main contribution is to construct square resultant matrices, which are submatrices of those introduced by Cattani et al. (1998), and whose determinants are nontrivial multiples of the sparse (or toric) resultant. The matrix is hybrid in that it contains a submatrix of Sylvester type and an additional row expressing the toric Jacobian. If we restrict our attention to matrices of (almost) Sylvester-type and systems as specified above, then the algorithm yields the smallest possible matrix in general. This is achieved by strongly exploiting the combinatorics of sparse elimination, namely by a new piecewise-linear lifting. The major motivation comes from systems encountered in geometric modeling. Descripción: The relationship between water transport and photosynthesis represents the trade-off between carbon gain and water loss and was used to evaluate potential differences in water resource utilization among two dominant vegetation types of south Florida: subtropical evergreen broad leaf forests (hardwood hammocks) and pine woodlands (pine rocklands). We found consistent linear positive relationships between the quantum yield of photosystem II (φPSII), an index of photosynthetic capacity, and hydraulic conductivity per sapwood area (kS) and per leaf area (kL) across all species. The slope of the φPSII-kS relationship was steeper for hardwood hammock than for pine rockland species. Mean φPSII was greater in pine rockland species and was greater for a given kL than in hardwood hammock species. These results are consistent with previous observations demonstrating that pine rocklands tend to have better access to stable water sources than hardwood hammocks. We also found greater photosynthetic carbon isotope discrimination with increasing kS and kL in pine rockland species, but not in hardwood hammock species, suggesting increased stomatal conductance with increasing kS and kL, consistent with greater water availability in pine rockland habitats. Our study thus utilizes relationships between water transport and photosynthesis to evaluate hydraulic constraints on physiological function between two contrasting vegetation types with contrasting stability of water sources. © 2010 The Author(s).

Descripción: A stationary, one-dimensional fluid model is presented to describe the interelectrode region of a nonfiltered vacuum arc operated with a background gas. The model includes the electron energy equation and the main elastic and inelastic atomic processes among metallic ions, electrons, and gas particles. To validate the model predictions an experimental study of the plasma-neutral gas structure, using a titanium (Ti) cathode and argon (Ar) as the background gas, is presented. The measured electron temperature and the experimental dependence on the pressure of neutral Ti and Ar spectroscopic emission lines are well reproduced, using a simple atomic model to interpret the plasma radiation emission. © 2006 American Institute of Physics.

Descripción: We present a simple hydrodynamic model to obtain the profiles of the relevant physical quantities along a nozzle of arbitrary cross-section in a cutting torch. The model uses a two-zone approximation (a hot central plasma carrying the discharge current wrapped by a relatively cold gas which thermally isolates the nozzle wall from the plasma). Seeking for a solution with sonic conditions at the nozzle exit, the model allows expressing all the profiles in terms of the externally controlled parameters of the torch (geometry of the torch, discharge current, mass flow of the gas and plenum pressure) and the values of the arc and gas temperatures at the nozzle entrance. These last two values can be estimated simply appealing to energy conservation in the cathode-nozzle region. The model contains additional features compared with previous reported models, while retaining simplicity. The detailed consideration of an arc region coupled to the surrounding gas dynamics allows determining voltage drops and consequent delivered power with less assumptions than those found in other published works, and at the same time reduces the set of parameters needed to determine the solution.

Descripción: A hydrodynamic numerical study at the mouths of the Paraná and Uruguay rivers and the upper Río de la Plata is presented in this paper. Water Quality Mapping numerical model was implemented and realistic and very simple boundary conditions were specially developed for this complex estuarial system. A set of numerical experiments were carried out using different constant discharges for the Paraná and Uruguay rivers but unrealistic currents were generated. In order to obtain more realistic results, a set of numerical simulations were carried out imposing water level timeseries at the open boundaries. M2, S2, K1 and O1 harmonic constants were used to generate water levels at Zárate (Paraná river), Nueva Palmira (Uruguay river) and the eastern boundary of the domain (La Plata-Colonia). A mean water level equal to zero was set between La Plata and Colonia. Positive mean water levels (0.3-0.4m) were imposed at Zárate and Nueva Palmira to simulate the hydraulic slope of both rivers and, consequently, to generate realistic and unsteady discharges. These boundary conditions, built by means of the addition of a mean water level and the astronomical tide, significantly improve the simulated currents at the northernmost region of the RDP estuary. © 2011 Elsevier Inc.

Descripción: The rapid changes that are taking place in the climate of the Antarctic Peninsula are triggering hydrological processes which had been limited or inactive for relatively long periods of time. These processes are evident in ice-free areas on the northern edge of the Antarctic Peninsula, such as Cape Lamb, and lead to intense draining of the surface and groundwater system throughout the brief Antarctic summer. The result is a movement of large amounts of water, sediments and nutrients that would be immobilized in other scenarios. This study proposes a model of the operation of the surface and groundwater system that could be valid for several ice-free areas in the region, based on the interpretation of field observations in Cape Lamb, the most extensive ice-free area on Vega Island. The model proposed is further supported by the interpretation of 56 chemical analyses of samples representing groundwater, active layer water, glacier ice and snowfall. The hydrochemical interpretation is supported primarily by four indicators (pH, electrical conductivity, D/18O ratio and SO4/Cl ratio) which have proven to be the most appropriate hydrochemical variables to differentiate the origin and interaction of the waters in the various sections of the system.

Descripción: Since the elucidation of the myoglobin (Mb) structure, a histidine residue on the E helix (His-E7) has been proposed to act as a gate with an open or closed conformation controlling access to the active site. Although it is believed that at low pH, the His-E7 gate is in its open conformation, the full relationship between the His-E7 protonation state, its conformation, and ligand migration in Mb is hotly debated. We used molecular dynamics simulations to first address the effect of His-E7 protonation on its conformation. We observed the expected shift from the closed to the open conformation upon protonation, but more importantly, noted a significant difference between the conformations of the two neutral histidine tautomers. We further computed free energy profiles for oxygen migration in each of the possible His-E7 states as well as in two instructive Mb mutants: Ala-E7 and Trp-E7. Our results show that even in the closed conformation, the His-E7 gate does not create a large barrier to oxygen migration and permits oxygen entry with only a small rotation of the imidazole side chain and movement of the E helix. We identify, instead, a hydrophobic site in the E7 channel that can accommodate an apolar diatomic ligand and enhances ligand uptake particularly in the open His-E7 conformation. This rate enhancement is diminished in the closed conformation. Taken together, our results provide a new conceptual framework for the histidine gate hypothesis. © 2013 by The American Society for Biochemistry and Molecular Biology, Inc.

Descripción: A result of Godefroy and Shapiro states that the convolution operators on the space of entire functions on Cn, which are not multiples of identity, are hypercyclic. Analogues of this result have appeared for some spaces of holomorphic functions on a Banach space. In this work, we define the space holomorphic functions associated to a sequence of spaces of polynomials and determine conditions on this sequence that assure hypercyclicity of convolution operators. Some known results come out as particular cases of this setting. Descripción: Although transcriptional effects of thyroid hormones have substantial influence on oxidative metabolism, how thyroid sets basal metabolic rate remains obscure. Compartmental localization of nitric-oxide synthases is important for nitric oxide signaling. We therefore examined liver neuronal nitric-oxide synthase-α (nNOS) subcellular distribution as a putative mechanism for thyroid effects on rat metabolic rate. At low 3,3′,5-triiodo-L-thyronine levels, nNOS mRNA increased by 3-fold, protein expression by one-fold, and nNOS was selectively translocated to mitochondria without changes in other isoforms. In contrast, under thyroid hormone administration, mRNA level did not change and nNOS remained predominantly localized in cytosol. In hypothyroidism, nNOS translocation resulted in enhanced mitochondrial nitric-oxide synthase activity with low O2 uptake. In this context, NO utilization increased active O2 species and peroxynitrite yields and tyrosine nitration of complex I proteins that reduced complex activity. Hypothyroidism was also associated to high phospho-p38 mitogen-activated protein kinase and decreased phospho-extracellular signal-regulated kinase 1/2 and cyclin D1 levels. Similarly to thyroid hormones, but without changing thyroid status, nitric-oxide synthase inhibitor Nω-nitro-L-arginine methyl ester increased basal metabolic rate, prevented mitochondrial nitration and complex I derangement, and turned mitogen-activated protein kinase signaling and cyclin D1 expression back to control pattern. We surmise that nNOS spatial confinement in mitochondria is a significant downstream effector of thyroid hormone and hypothyroid phenotype. © 2006 by The American Society for Biochemistry and Molecular Biology, Inc.

Descripción: Multilevel resistance states in silver-manganite interfaces are studied both experimentally and through a realistic model that includes as a main ingredient the oxygen vacancies diffusion under applied electric fields. The switching threshold and amplitude studied through hysteresis switching loops are found to depend critically on the initial state. The associated vacancy profiles further unveil the prominent role of the effective electric field acting at the interfaces. While experimental results validate main assumptions of the model, the simulations allow to disentangle the microscopic mechanisms behind the resistive switching in metal-transition metal oxide interfaces. © 2010 American Institute of Physics.
